  In vitro collection of valuable genotypes of deciduous woody plants (hereafter – the Collection) was founded in 1991

  The Collection is registered in the All-Russian Research Institute of Forest Genetics, Breeding and Biotechnology (russian abbreviation — VNIILGISbiotech, hereafter – the Institute) on the 10th of April, 2014 by order of the director № 8/2-П.

  The Collection was formed for in vitro (or ex situ) conservation and reproduction (cloning) of valuable and exclusive genotypes of deciduous woody plants including species in danger of extinction and/or those that do not respond well to vegetative reproduction. Among other things the Collection can be used for basic and task-oriented research works in the field of forest genetics, breeding and biotechnology. The Collection was one of the first in Russia (created in 1991), and is unique thanks to its composition and the duration of in vitro storage (over 26 years) of the samples. As a comparison, the duration of storage of rare species of Betulaceae family at the Forest Research Department of the Karelian Branch of Russian Academy of Sciences (Petrozavodsk) is 14 years. As at the January, 2018 the Collection of the Institute has over 60 clones of deciduous woody microplants (including 18 species, 3 genera and 2 families). Among them are economically valuable and exclusive clones, hybrids, polyploids and varieties of birch, poplar, aspen and willow, either naturally selected or produced by breeding. Every year we enrich the Collection with new genotypes and species, sometimes swapping samples with other relevant organizations through cooperation with a range of research institutes in Russia.

   The long-term maintenance of clones is possible due to new methods developed by the Institute. The first method takes place under normal growth conditions in which microplants are subjected to rare subcultivation on hormone-free medium (with the exception of willow clones) under normal (standard) climate conditions. The second method takes place under inhibited growth conditions in which microplants are subjected to in vitro deponation at low temperature in the refrigerator and media contain substances that inhibit the growth).

   The Institute has a successful experience of creation of purpose-oriented forest plantations of birch, poplar and aspen trees grown on the basis of the Collection, the current age of those trees varies between 5 and 26 years.

  The long-term maintenance and development of the Collection is done within the framework of the state programme "BIO2020", the implementation of the roadmap for "Development of biotechnology and gene engineering", which has been approved by the government of the Russian Federation on the 18th of July, 2013, № 1247-р and the establishment of the Forest Biotechnology Center.

The principal lines of the scientific activity carried out with the use of the Collection are:

  • in vitro (ex situ) conservation and reproduction (cloning) of economically and scientifically valuable genotypes of deciduous woody plants;
  • improvement and unification of technological methods of long-term in vitro storage;
  • development of new methods and techniques of in vitro cloning of valuable genotypes that do not respond well to vegetative reproduction;
  • expanding of the Collection with new samples;
  • creation of an information database of clones, including their certification and field testing results;
  • basic and task-oriented research works in the field of biotechnology, genetics, cytology, anatomy, developmental biology, breeding; as well as research on cell and tissue selection for improvement of existing forms and obtaining new forms.
